چکیده

In a previous paper (1) evidence was presented that acid hydrolysates of the cellular material of BaciZZus brevis (ATCC 10068) contained an unusually high content of amino acids of the D configuration, amounting to over 10 per cent of the total amino acids or 5 per cent of the dry weight of the cells. The n-amino acids were shown to exist in the intact cells, and not to result from racemization during acid hydrolysis, by feeding the cells to rats and measuring the n-amino acids excreted in the urine. Studies of the nature of antibiotic peptides produced by various strains of this organism (cf. (2, 3)) have demonstrated that at least two of the compounds are cyclic decapeptides containing one or more amino acid residues of the D configuration. It seemed, therefore, of interest to survey the entire amino acid composition of the cells of B. brevis to determine quantitatively the content of the various amino acids of the D configuration, and, if possible, to determine the mode of occurrence of the n-amino acids in the intact cells.

Use of both translation initiation sites of the middle wall protein gene in Bacillus brevis 47.

The middle wall protein gene of Bacillus brevis 47 has two potential translation initiation sites located tandemly in the same reading frame. We demonstrate here that both sites are utilized to start translation in B. brevis 47. Translation from the first site (located upstream) gives rise to a precursor of the middle wall protein with an extension peptide of 31 amino acids preceding the signal...

THE OCCURRENCE OF d-AMINO ACIDS IN GRAMICIDIN AND TYROCIDINE

Amino acids having a dextro configuration have been found in natural material from two sources.’ Jacobs and Craig obtained d-proline from ergotinine (4) and later other ergot alkaloids, and IvAnovics and Bruckner found d( -)-glutamic acid as the principal constituent of the P-antigen of Bacillus anthracis and other microorganisms of the related Bacillus mesentericus-subtilis group (5). Inasmuch...

Efficient synthesis and secretion of a thermophilic alpha-amylase by protein-producing Bacillus brevis 47 carrying the Bacillus stearothermophilus amylase gene.

Bacillus subtilis and Bacillus brevis 47-5, carrying the Bacillus stearothermophilus alpha-amylase gene on pUB110 (pBAM101), synthesized the same alpha-amylase as the donor strain as determined by the enzyme's thermal stability and NH2-terminal amino acid sequence. Regardless of the host, the 34-amino acid signal peptide of the enzyme was processed at exactly the same site between two alanine r...

Purification and properties of branched chain amino acid aminotransferase from gramicidin S-producing Bacillus brevis.

The branched chain amino acid aminotransferase [EC 2.6.1.42] was purified to a homogeneous state from a gramicidin S-producing strain of Bacillus brevis. The enzyme had a molecular weight of about 93,000 and consisted of two identical subunits, each with a molecular weight of about 47,000. One pyridoxal phosphate is bound per subunit. In addition to branched chain amino acids, the enzyme uses L...
